This is a cloned mast cell line derived from the fetal liver of a (B6 X A/J)F1 mouse.

Comments
The cells can be sensitized to specific antigens by incubating them with IgE having the desired antigenic specificity.

The cells so coated will release soluble mediators (such as histamine) in response to exposure to the antigen (allergen).
Complete Growth Medium Dulbecco's Modified Eagle's Medium with 4 mM L-glutamine adjusted to contain 4.5 g/L glucose and 1.5 g/L sodium bicarbonate and supplemented with 2 mM L-glutamine, 0.05 mM 2-mercaptoethanol, 10% Rat T-STIM (Becton Dickenson Catalog No. 354115) and 10% fetal bovine serum.
Subculturing
Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 to 5 X 105 viable cells/ml. Maintain cell density between 2 X 105 and 2 X 106 viable cells/mL. Do not allow the cell density to exceed 2 X 106 cells/mL.
